I have a 2014 JKUR with a manual and 4.10's, I even have 35" STT's but not the pro, I wish I had 4.56 on the highway, but I live in a hilly area.

I'm not sure of the overdrive ratio differences between the auto and manual..

'15 JKUR auto here with 35's and 4.10's - I've been happy with the combo. No issues on the highways, passing, gear hunting, etc... Have a friend with a '15 2dr auto, 35's, on 3.73 - and its actually also quite satisfactory (on and offroad). Both in Michigan where highways are typically flat though.

If you know 35's are what you're sticking with, I'd go 4.10; I don't think I'd consider 4.56, unless perhaps you think you might want to go 37's later on.

I'd imagine 4.56's will be better, especially if you're thinking to add extra weight later on.

Personally, I would go 4:88s. in fact I just went 5:13s in anticipation of 37s on a 14 JKUR auto and I love the way it runs. Night and day difference from 4:10s in an auto. I never saw overdrive with 4:10s and 35s........

Hell I've got 4.56 on my '13 auto sport with 33's (285 70 17's) and I wouldn't want anything different, especially if I were considering bigger tires. Love the grunt with these gears

I have 2014 JKUR manual on 35s as well, Swapped 4.10s for 4.56. Got me back to stock rubi ratio.

Here is a handy calculator.. That will give you info on rpm and speed, based on gears and transmission.. Allows you to do side by side comparison

I have 4.56 gears with 33's. 2014 manual... I love it. I am going up to 35's or 37's if these tires ever wear out though. I will say that gear ratio is great with 33's, i'm at about 2600-2700 rpm's running 65-70
